人白介素-2（IL-2）ELISpot试剂盒用于对外周血单个核细胞（PBMCs）悬液中分泌人IL-2的细胞进行定量检测和计数。ELISpot常用于评估特异性免疫反应，如确定对抗原刺激产生反应的活化T细胞亚群的占比。IL-2主要由活化的CD4+ T细胞分泌，部分由CD8+ T细胞分泌，是T细胞生长和发育的重要调节因子。ELISpot具有高灵敏度，可在单细胞水平检测分泌的分析物。该检测用于传染病、肿瘤、过敏和自身免疫性疾病研究。

本检测基于改良的夹心ELISA方法，将细胞加入预包被有该细胞因子特异捕获抗体的ELISpot板中。细胞因子在整个培养期间被捕获，并通过加入生物素化检测试剂抗体检测，随后加入链霉亲和素-碱性磷酸酶（SA-ALP）与生物素化抗体结合。加入显色酶底物5-溴-4-氯-3-吲哚基-1-磷酸（BCIP）/硝基蓝四唑（NBT）后形成有色斑点。这些斑点的数量、大小和强度与样品中细胞因子分泌细胞的百分比及分泌的分析物量成正比。

ELISpot analysis of human peripheral blood mononuclear cells (PBMCs) isolated from whole blood using EasySep™ Direct Human PBMC Isolation Kit (Catalog #19654). Cells were cultured in ImmunoCult™-XF T Cell Expansion Medium (Catalog #10981) in the absence of stimuli (negative control) or presence of Anti-human CD3 mAb (CD3-2), azide free (included in kit) or ImmunoCult™ Human CD3/CD28/CD2 T Cell Activator (Catalog #10970). Cells were seeded at 5 x 10^4 cells/well on IL-2 pre-coated white plate and incubated at 37°C, 5% CO2 for 18 hours. Following incubation, ELISpot plates were processed as per the instructions on the Product Information Sheet. Wells were imaged and counted using an ELISpot reader.
